SINGAPORE – 8 Jul 2024 – Tikva Allocell Scientists published scientific article in Cell Molecular Therapy on Novel OX40 and 4-1BB Derived Spacers Enhance CD30 CAR Activity and Safety in CD30 Positive Lymphoma Models.
The chimeric antigen receptor is made up of several domain, all of which have important roles to play in determining the safety and efficacy of the CAR. However, a domain that is commonly overlooked in CAR engineering is the spacer domain. The length and flexibility of the spacer domain critically affects the ability of the CAR to reach and bind the target epitope. Furthermore, careful selection of the spacer domain will ensure undesirable interactions that potentially impact the CAR activity negatively.
In our recent publication, we show the use of a novel spacer domain derived from 4-1BB and OX40 costimulatory receptors in achieving potent killing of tumors while minimizing secretion of proinflammatory cytokines that would compromise safety.
Thus, guided by a good understanding of the epitope recognized by the antigen binding domain, we demonstrate that the spacer domain can be rationally designed to enhance performance and developability.
